Ectonucleoside triphosphate diphosphohydrolase-1 (gene: ENTPD1; protein: NTPDase1) also known as CD39 (Cluster of Differentiation 39), is a typical cell surface enzyme with a catalytic site on the extracellular face.

Here, we first review the environmental and genetic factors shaping CD39 expression. Second, we report CD39 functions in the T cell compartment, highlighting its role in regulatory T cells, conventional CD4 + T cells and CD8 + T cells.

In this review, we provide a background on the molecular regulation of CD39 and CD73, outline the role of CD39 and CD73 in governing immune cell function, and detail the role of these enzymes in regulating the course of immune-mediated diseases.
